A Post-doctoral Fellow position for a highly motivated researcher is available in Prof. Merari F. R. Ferrari Laboratory at the Institute of Biosciences of the University of São Paulo (campus São Paulo).
Our Laboratory is interested in studying cell biology of neurodegenerative diseases.
The focus of the work will be a combination of approaches including: cell culture (primary, fibroblast and stem cell), in vivo analysis, cloning, neuron physiology, molecular and cell biology of neuron, as well as protein aggregation related with neurodegeneration.

About the project
Post-doctoral fellow will establish dopaminergic neurons derived from iPS of PD patients as well as from controls. Epidermal fibroblasts from patients diagnosed with Parkinson's disease as well as from controls were purchased from the Coriell Institute and reprogrammed to hiPS cells. These cells will be characterized and differentiated into midbrain dopaminergic neurons. The Postdoc fellow will also establish primary cultures of hippocampal neurons from AD transgenic mice.
These cells will be subjected to the analysis of intracellular dynamics, trafficking and cross-talk between ER and Mitochondria. To detect mitochondrial movement and autophagic vacuoles movement he will use live-cell imaging using MitoTracker Green FM (Invitrogen) and using Cyto-ID™ Autophagy detection kit (Enzo, Lausen, Switzerland), respectively.
To study Microtubules (MT)-dependent ER movements, cells will be co-transfected with GFP-tubulin (to visualize MT) and pDSRed2-ER (to visualize ER dynamics), on gridded coverslips. Microscopic evaluation will be complemented with quantification of protein levels, by Western blot, of STIM1, EB1 and CLIMP63 (involved in MT-mediated ER dynamics) and atlastin-1 (regulates ER fusion).
To study ER-mitochondria interaction cells will be co-transfected with mito-GFP (to visualize mitochondria) and pDSRed2-ER (to visualize ER). ER-mitochondria contacts will be also analyzed through electron microscopy. Transfer of calcium between the ER and mitochondria plays a key role in the life of numerous cell types and it is disrupted in several neurodegenerative disorders, namely AD. Ca2+ shuttling from ER to mitochondria will be carried out using fluorescent probes as Fura-2AM (Sigma) and a motorized Zeiss LSM 510 confocal microscopy Meta/UV microscope with Axiovert 200.
The next set of experiments will be performed in order to evaluate starvation effect on protein aggregation, mitochondrial and ER function, dynamics, trafficking and cross-talk in iPS-derived neurons and neurons from AD transgenic mice, techniques to measure these parameters were as detailed in the previous paragraph.
Contract period and project Start
The scholarship duration is 30 months, starting in March 1st, 2019.
Values and conditions
1. Monthly grant (free of taxes) of BRL 7,373.10, plus 15% of the annual value for expenses related to the research (Technical Reserve);
2. Financial support for travel and installation expenses may be requested for the selected applicant who lives out of São Paulo and the merit will be evaluated by FAPESP upon acceptance of the stipend;
3. The candidate must have completed the PhD no longer than three years prior to application;
4. The grantee must not have any formal or informal employment, nor receive during the stipend a fellowship of another agency, salary or remuneration due to his/her activities of any nature.
